New evidence suggests that the survival rate in patients with metastatic non-small cell lung cancer (NSCLC) does not significantly improve with the addition of the antibody-drug conjugate (ADC) sacituzumab govitecan (Trodelvy) to standard immunotherapy treatments, according to findings from a large randomized trial.
Study Insights
The EVOKE-03 trial reported a median progression-free survival (PFS) increase from 7.7 months with pembrolizumab (Keytruda) alone to 11.8 months when combined with sacituzumab. However, this difference did not meet the predefined criteria for statistical significance. An interim analysis of overall survival (OS) showed a slight benefit for the control group (22.8 vs 21.5 months), particularly in a population selected for PD-L1 positivity. Additional analyses from patients in East Asia and China indicated potential OS advantages for the sacituzumab group, but these findings were not statistically significant.
Treatment-related Adverse Events
Adverse events associated with treatment, including serious and fatal outcomes, occurred more frequently in patients receiving sacituzumab, as reported by Dr. Giannis Mountzios from Henry Dunant Hospital Center in Athens, Greece, during the World Conference on Lung Cancer (WCLC) in Seoul, South Korea.
