Eficacia en Cáncer de Pulmón
The oral RAS inhibitor daraxonrasib (Rasonque) has demonstrated efficacy in patients with metastatic RAS-mutant non-small cell lung cancer (NSCLC) who have previously undergone treatment. This was revealed in a phase I/II trial involving 136 patients, where the objective response rate varied with dosage: 31% for 120 mg or less, 34% for doses between 160 and 220 mg, and 37% at 300 mg, as reported by Kathryn C. Arbour, MD, at Memorial Sloan Kettering Cancer Center.
Eventos Adversos
Grade 3 or higher adverse events (AEs) were reported in 54% of the patients, including pneumonia (10%), diarrhea (9%), rash (8%), and anemia (5%). Notably, four grade 5 adverse events were observed, highlighting the importance of monitoring during treatment.
Mutaciones en RAS
Mutations in the RAS gene are common in NSCLC, affecting around 30% of patients. Unfortunately, there are currently no approved targeted therapies specifically for these mutations.
FDA y Aprobación
Daraxonrasib has recently received FDA approval for metastatic pancreatic ductal adenocarcinoma, marking a significant achievement based on the results of the RASolute 302 trial, which indicated that it doubled median overall survival in patients with specific RAS G12 mutations.
Estudios de Eficacia
In an evaluation of docetaxel-naive patients who had received other treatments, daraxonrasib showed a confirmed objective response rate of 42% after a median follow-up of 18.3 months, with a median progression-free survival of 8.3 months and a median overall survival of 16 months.
Consideraciones de Dosis
The study noted that patients receiving doses between 160 and 220 mg experienced a 51% rate of grade 3 or higher AEs, leading to dose modifications in 71% of cases. This suggests the need for optimal disease-specific dosing.
Conclusiones y Advertencias
While the results are promising, they should be interpreted with caution, given the phase I/II trial design and small sample sizes within RAS mutation categories. Further studies will be necessary to provide more definitive conclusions and outcomes.
